Installing Puppy Linux on a USB flash is very easy and I always run my Puppy Linux only from USB.
I usually install my Puppy linux to USB use a very usefull application it is Unetbootin. I do this in Ubuntu but you can run Unetbootin from windows operating system.
Here are what I do to install Puppy Linux to USB.
Step One
Download the latest Puppy Linux release from Puppylinux.org the .iso is very small it is around 125 MB and save in your computer.
Step Two
Download and run Unetbootin. In ubuntu, just run sudo apt-get install unetbootin in terminal. For windows OS just download it from Unetbootin site and run it.
Browse Puppy Linux iso in your computer and then click OK button to process and wait a minutes.
Step Three
Restart or Reboot your computer and set your bios to boot from USB then restart your computer then you can run puppy linux from USB.
If you want to keep using your USB make sure to make save file at first shut down and save your puppy personal save in to your USB or hard disk. This save file will save all of your data configurations and settings include your additional applications if you install some applications. And every time boot, puppy will load your save file.

This way will not work if your computer does not support booting from USB, so the alternative is to burn puppy iso to CD using burning software.
